QuadMix is administered as a penile self-injection, typically considered to be the most powerful class of anti-erectile dysfunction agents.1 While the components of QuadMix (Papaverine, Phentolamine, Prostaglandin E1, Atropine) are, on their own, indicated for a vast number of different conditions, the practice of bringing them together in concert to treat erectile dysfunction has become commonplace in sexual medicine21 and is now considered the go-to treatment if a patient is not responsive to conventional PDE5 inhibitors such as Viagra or Cialis.1 QuadMix is used in the treatment of erectile dysfunction in males. QuadMix contains four drugs from complimentary classes designed to act synergistically, mixed into a sterile injection. They are:

Papaverine: Blocks the breakdown of cAMP and cGMP (cyclic Guanosine Mono-Phosphate), leading to a decrease in intracellular calcium and the relaxation of smooth muscle that may result in and has been reported to increase duration of erection.

Phentolamine: A non·selective Alpha Blocker adjunct therapy that inhibits smooth muscle contraction and has a direct effect on smooth muscle activity. Phentolamine has been reported to provide a synergistic effect when combined with PGEI and Phentolamine.

Prostaglandin E1: Directly stimulates the cAMP (cyclic Adenoslne Mono-Phosphate) pathway to decrease intracellular catdum concemraaons, allowing for relaxation of arterial and smooth muscle tissue that may result in and has been reported to increase firmness.

Atropine: A parasympatholytic drug that enables the relaxation of smooth muscle tissue that is derived from the release of endothelium. This may result in and has been reported to increase firmness when used as an adjunct therapy.
